The organization and polarity of pollen mother cells of Triticum aestivum.

چکیده
Colchicine has been applied to young developing anthers of Triticum aestivtim at varying stages of maturity from the last premeiotic mitoses of the archesporial and tapetal cells to the second meiotic divisions of the pollen mother cells. The developmental stage of the archesporium at which colchicine took effect was determined by cytological examination of the ploidy levels of the nuclei of the adjacent tapetal cells. The type of pollen abnormality induced depended on the time of application and the concentration of colchicine. Uninucleate monads with 4 randomly positioned pores and uninucleate monads without pores were obtained with 0 5 % colchicine. Multipored polyads and multipored uninucleate monads were observed together in anthers treated with o-oi % colchicine. Naturally occurring aberrant pollen types in hybrids of Triticum aestivum x Aegilops mutica or T. aestivum x Aegilops sharonensis have revealed a constant relationship between the disposition of the meiotic spindles and the siting of the pollen pores. The colchicine-induced abnormalities have further clarified the nature of this relationship leading to the interpretation that both the positioning of the spindles and the siting of the pores are predetermined by events taking place in the premeiotic interphase at a time just after the last mitosis of the pollen mother cells and the penultimate mitosis of the tapetum. A reorganization of the archesporial cells (sensitive to colchicine) possibly occurs at this time. Various subsequent meiotic events are dependent on the reorganization. Two of these events the organization of meiotic spindles and the establishment of pollen symmetry are discussed.

The effect of spindle inhibitors applied before meiosis on meiotic chromosome pairing.
Injection of 0-5 % colchicine into immature tillers of genotypes of Triticum aestivum, T. aestivum x Aegilops mutica and T. aestivum x Secale cereale hybrids induces asynapsis at first meiotic metaphase irrespective of the homologous or homoeologous nature of the potential pairing chromosomes. The induction of asynapsis occurs at a time during and immediately following the last premeiotic mitos...
متن کاملMicrotubules and root protophloem ontogeny in wheat.
Protophloem ontogeny in roots of Triticum aestivum has been investigated ultrastructurally. Each protophloem pole consists of three cells, a protophloem sieve element and two companion cells, all originating from a single precursor cell usually having a pentahedral shape. This protophloem mother cell (PMC) undergoes two successive asymmetrical divisions: the first one gives rise to a smaller ce...
متن کاملRelationship between male sterility and β-1,3-glucanase activity and callose deposition-related gene expression in wheat (Triticum aestivum L.).
In previous studies, we first isolated one different protein β-1,3-glucanase using two-dimensional electrophoresis and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry from normal wheat (Triticum aestivum L.) and chemical hybridization agent-induced male sterility (CIMS) wheat. In this experiment, β-1,3-glucanase activity and the expression of a callose deposition-re...
متن کاملUltrastructural studies on the sporogenous tissue and anther wall of Leucojum aestivum (amaryllidaceae) in different developmental stages.
In this study, ultrastructures of anther wall and sporogenous tissue of Leucojum aestivum were investigated during different developmental stages. Cytomictic channels were seen between pollen mother cells during prophase I. Polar distribution was described in the organelle content of pollen mother cells and microspores in early phases of microsporogenesis and also in pollen mitosis. Active secr...
متن کاملQTug.sau-3B Is a Major Quantitative Trait Locus for Wheat Hexaploidization
Meiotic nonreduction resulting in unreduced gametes is thought to be the predominant mechanism underlying allopolyploid formation in plants. Until now, however, its genetic base was largely unknown. The allohexaploid crop common wheat (Triticum aestivum L.), which originated from hybrids of T. turgidum L. with Aegilops tauschii Cosson, provides a model to address this issue. Our observations of...
